Study on the sealing performance of ampoules for freeze-dried bacterial strains

There are certain requirements for the placement tube of freeze-drying bacteria, and the size of the tube depends on the diameter of the conical short tube on multiple branches. We choose an inner diameter of 8 m and a glass tube of 110 mm. The quality requirements for glass are neutral, and alkaline glass cannot be used. Their texture and hardness have advantages and disadvantages. Soft glass is easy to melt and open during recovery and cultivation, but it is easy to break during storage. Glass with high hardness is not easy to melt or open, but it hardly breaks during storage. The company can order from Glass production according to actual needs. When selecting ampoules, use 2% first; Soak the robe with HCl solution 8 × 10 hours, then rinse repeatedly with tap water. Wash with distilled water 2-3 times and finally air dry.
Vacuum sealing, no matter which type of freeze-drying equipment is used, the sealing method of ampoules by the ampoule melting and sealing machine is the same. In a vacuum state, the upper end of the ampoule is melted, forming a thin neck for quick sealing. It is then installed on the side of multiple collecting pipelines, and the vacuum pump is started. When the true vacuum is 0.001 torr, the seal is applied at the thin neck. If the vacuum cannot reach such low pressure, then it should be adjusted to at least 0.01 torr. Only by storing freeze-dried bacteria in such a high vacuum environment can ideal results be achieved. After the ampoule is sealed, a high-frequency electric spark generator should be used for testing. If a light purple or white electric spark appears in the ampoule, it indicates that it is still in a vacuum state. If a blue or purple spark appears, it indicates that the vacuum has been lost.
