1. Ampoule preparation method: Pour purified water into the ampoule, heat at 100 ℃ for 30 minutes, and then throw out when hot. Then, pour the filtered purified water and injection water into the ampoule, and throw out the water three times to remove free alkalis, metal ions, dust, and attached sand from the surface of the ampoule. After cleaning, place at 120-140 ℃ for use.
2. Process of vertical melting glass filter: clean the vertical melting glass filter with pure water, soak it with 1%~2% Sodium nitrate sulfuric acid solution for 12-24 hours, and then repeatedly wash it with pure water and injection water until the solution is neutral and transparent, then take it out for use.
3. Clean and dry the prepared liquid and liquid preparation equipment according to the specified requirements. There are usually two ways to prepare liquids.
(1) Dilution method: Add the raw material to a solvent at once to achieve the required concentration.
(2) Concentration method: Mix the raw materials with certain solvents to form a concentrated solution, then heat and filter. If necessary, add activated carbon or freeze, then filter and dilute to the desired concentration.
4. The filtering methods for filtration include pressure filtration, reduced pressure filtration, high-pressure static pressure filtration, etc. There are various types of filters used for coarse filtration, pre filtration, and fine filtration. Set up filtration equipment according to the laboratory environment.
5. Pour the filtered solution into the seal and seal it with an ampoule melting machine. The dosage should be precise and should not touch the neck side. Easy oxidizer, which can allow inert gas to pass through during filling.
6. Disinfection and leakage disinfection and inspection shall be carried out according to the requirements after using a fusion sealing machine for fusion sealing. After disinfection, remove it and place it in 1% ice water for testing.
7. The aqueous solution (liquid) used for intravenous infusion should be free of pyrogens. Unless it meets the requirements of general injections, insoluble particles must comply with regulations and should be as permeable as possible to the blood. For lotion for intravenous infusion, the purity of its dispersed phase pellets is 80%, less than 1 μ m, no particles more than 5 μ m, no pyrogen, resistant to high temperature sterilization, stable during storage, and cannot be used for spinal canal injection. In addition, the pH value used for infusion should be consistent with the pH value of the human body as much as possible, and antibacterial agents should not be added, nor should they cause abnormalities in blood count. The preparation method is basically the same as ampoules.
